Laboratory for Large Scale Organic Synthesis

The Laboratory for Large Scale Organic Synthesis was established in 2024 to enable the synthesis of large quantities of research material, precursors for natural product synthesis and drug candidates for preclinical investigations and formulation.

The laboratory is equipped with large walk-in fume hoods to enable the safe operation of the following instrumentation:

  • 50 liter reactor (+150°C to -50°C)
  • 20 liter reactor (+150°C to -50°C)
  • 20 liter filter reactor (+150°C to -50°C)
  • 50 liter rotary evaporator
  • 5 liter rotary evaporator

Large scale purification is supported by distillation, recrystallization, and column chromatography. We have a 5L setup for fractional vacuum distillation. Recrystallizations can be carried out in a 20L filter reactor followed by drying using our large vacuum and gravimetric ovens. Our automated column chromatography system from Biotage (Isolera LS) can be operated with up to 5 kg of column material such as silica, aluminum oxide or C18 to enable purification of up to 100 g of material in one run.

Reaction monitoring and purity analysis are carried out using our LCMS 2020 (Shimadzu) . The system has Dual Ion Source DUIS that simultaneously ionizes samples using two ionization methods – electrospray ionization (ESI), which is suitable for high polarity compounds, and atmospheric pressure chemical ionization (APCI), which is suitable for low-to-medium polarity compounds. Furthermore, the UF switch enables positive and negative ionization at the same time. Of course the system is equipped with a PDA detector for purity analysis.

The facility also houses high pressure reactors to support hydrogenations and other reaction that uses gases and gas mixtures. Currently, we have the following reactors available:

  • 100 mL liter reactor (up to 200 bar and +200°C)
  • 1 liter reactor (up to 200 bar and +200°C)
  • 5 liter reactor (up to 200 bar and +200°C)
