Pharmacokinetic Studies of ARN from Rat Plasma of GBM Xenografts

Letters & Science (College of) / Chemistry & Biochemistry

Project Description

Objective:
To analyze the pharmacokinetic properties of ARN (anti-tumor drug) in rat plasma, to establish an efficient drug for human glioblastoma.

Methodology:
1. Prepare the standard ARN, internal standard and rat plasma by performing solid phase extraction to extract the analyte from its matrix.
2. Identify and develop multiple reaction monitoring (MRM) method for both ARN and internal standard compound using LC-MS/MS.
3. Develop the calibration curve for the ARN.
4. Using the MRM method develop, analyze the concentration of ARN from the rat plasma in GBM xenograft.
5. Based on the concentration of ARN in plasma at particular time point, compute the pharmacokinetic properties of ARN from rat plasma in GBM xenograft.

Tasks and Responsibilites

1. Helping graduate student in identifying and developing multiple reaction monitoring (MRM) method for LC-MS/MS method.
2. Assisting in sample preparation for the generation of calibration curve.
3. Performing solid phase extraction using rat plasma.
4. Analyzing the concentration of ARN from rat plasma in GBM xenograft.
5. Computing the pharmacokinetic parameters for ARN in rat plasma.
