Project Description
Objective:
To perform and optimize the xenotransplantation of human GBM cells in zebrafish embryos at 24 hour post fertilization (hpf). The zebrafish model is used to study the efficiency of acid ceramidase inhibitors for treating GBM.Methodology
1. Dechorionate and mount the 24 hpf embryos.
2. Microinject the GBM cells into the hind brain of the embryos.
3. Monitor for recovery tumor growth of the zebrafish using optical and fluorescence microscope.
4. Introduce the acid ceramidase inhibitors to the zebrafish and monitor the tumor size and the cancer cell counts.
5. Perform toxicity assay of the acid ceramidase inhibitors in zebrafish larvae model.
Tasks and Responsibilites
1. Helping graduate student in optimizing the xenotransplantation of GBM in 24 hpf embryos.
2. Assisting in embryo dechrionation of the embryos and microinjection of GBM cells into the embryos.
3. Analyzing the tumor growth and its behavior towards acid ceramidase inhibitor using optical and fluorescence microscopes.
4. Computing the efficiency of the acid ceramidase inhibitors on GBM in zebrafish model.
