{"id":1176,"date":"2024-10-17T17:28:40","date_gmt":"2024-10-17T22:28:40","guid":{"rendered":"https:\/\/uwm.edu\/drug-discovery\/?page_id=1176"},"modified":"2026-02-25T11:11:25","modified_gmt":"2026-02-25T17:11:25","slug":"laboratory-for-large-scale-organic-synthesis","status":"publish","type":"page","link":"https:\/\/uwm.edu\/drug-discovery\/laboratories\/laboratory-for-large-scale-organic-synthesis\/","title":{"rendered":"Laboratory for Large Scale Organic Synthesis"},"content":{"rendered":"\n<div class=\"uwm-l-row\">\n<div class=\"uwm-l-col\">\n<p>The Laboratory for Large Scale Organic Synthesis was established in 2024 to enable the synthesis of large quantities of research material, precursors for natural product synthesis and drug candidates for preclinical investigations and formulation.<\/p>\n\n\n\n<figure><img loading=\"lazy\" decoding=\"async\" width=\"750\" height=\"463\" src=\"https:\/\/uwm.edu\/drug-discovery\/wp-content\/uploads\/sites\/390\/2024\/01\/Lab-for-large-scale-organic-synthesis-MIDD-2024.jpg\" alt=\"A large-scale organic synthesis laboratory featuring distillation columns, rotary evaporators, jacketed glass reactors, and other chemical processing equipment on open shelving.\" class=\"wp-image-1178\" srcset=\"https:\/\/uwm.edu\/drug-discovery\/wp-content\/uploads\/sites\/390\/2024\/01\/Lab-for-large-scale-organic-synthesis-MIDD-2024.jpg 750w, https:\/\/uwm.edu\/drug-discovery\/wp-content\/uploads\/sites\/390\/2024\/01\/Lab-for-large-scale-organic-synthesis-MIDD-2024-300x185.jpg 300w\" sizes=\"auto, (max-width: 750px) 100vw, 750px\" \/><\/figure>\n\n\n\n<p>The laboratory is equipped with large walk-in fume hoods to enable the safe operation of the following instrumentation:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>50 liter reactor (+150\u00b0C to -50\u00b0C)<\/li>\n\n\n\n<li>20 liter reactor (+150\u00b0C to -50\u00b0C)<\/li>\n\n\n\n<li>20 liter filter reactor (+150\u00b0C to -50\u00b0C)<\/li>\n\n\n\n<li>50 liter rotary evaporator<\/li>\n\n\n\n<li>5 liter rotary evaporator<\/li>\n<\/ul>\n\n\n\n<p>Large scale purification is supported by distillation, recrystallization, and column chromatography. We have a 5L setup for fractional vacuum distillation. Recrystallizations can be carried out in a 20L filter reactor followed by drying using our large vacuum and gravimetric ovens. Our automated column chromatography system from Biotage (Isolera LS) can be operated with up to 5 kg of column material such as silica, aluminum oxide or C18 to enable purification of up to 100 g of material in one run.<\/p>\n\n\n\n<p>Reaction monitoring and purity analysis are carried out using our LCMS 2020 (Shimadzu) . The system has Dual Ion Source DUIS that simultaneously ionizes samples using two ionization methods \u2013 electrospray ionization (ESI), which is suitable for high polarity compounds, and atmospheric pressure chemical ionization (APCI), which is suitable for low-to-medium polarity compounds. Furthermore, the UF switch enables positive and negative ionization at the same time.
